Refractometer รีแฟรกโตมิเตอร์

• ปริมาณน้ำตาล ในน้ำคั้นจากผลไม้สด น้ำเชื่อม เครื่องดื่ม เช่น ค่าที่ได้มีหน่วยเป็น องศาบริกซ์ (เปอร์เซ็นต์น้ำหนักต่อน้ำหนัก)
• การ ใช้ refractometer วัดน้ำคั้นจากผัก ผลไม้ หรือน้ำผลไม้ เป็นการวัดค่า total soluble solids หรือปริมาณของแข็งที่ละลายได้ทั้งหมด มีหน่วยเป็นเปอร์เซ็นต์ (เปอร์เซ็นต์น้ำหนักต่อปริมาตร)  เพราะในน้ำผลไม้ ประกอบด้วยน้ำตาลชนิดต่างๆ ซึ่งละลายได้ในน้ำ เช่น กลูโคส (glucose) ฟรักโทส (fructose) ซูโครส (sucrose) นอกจากน้ำตาแล้ว ในน้ำผลไม้ยังมีกรดอินทรีย์ ที่ละลายในน้ำได้ดี เช่น กรดซิตริก (citric acid) กรดมาลิก (malic acid) กรดทาร์ทาริก (tartaric acid) กรดแอมิโน และวิตามินซี ซึ่งค่าที่วัดได้ เป็นค่ารวมของความเข้มข้นน้ำตาลทุกชนิด และกรดอินทรีย์ที่ละลายได้ในน้ำผลไม้นั้น
• ปริมาณเกลือ มีชื่อรียกว่า Salinity Refractometer มีหน่วยเป็น ความเข้มข้น ppt (part per thousond, ถ้าต้องการค่าเป็น เปอร์เซ็นต์ ให้เอาค่า ptt/10) หรือ ความถ่วงจำเพาะของสารละลายเกลือ ใช้เตรียมน้ำเกลือในอาหารหมักเกลือ (salt curing) วัดความเข้มข้นของเกลือในซีอิ้ว น้ำปลา ปูเค็ม
• ปริมาณแอลกอฮอล์ ในเครื่องดื่มแอลกอฮอล์ (alcoholic beverage) มีหน่วยเป็น Alcohol by Volume หรือ % ABV
• ความหนาแน่นของของเหลว (liquid density) และ ความถ่วงจำเพาะของสารละลาย

หลักการทำงานของ Refractometers

Refractrometer ถูกประดิษฐ์ขึ้นมาโดย Dr. Ernst Abbe นักวิทยาศาสตร์ เยอรมัน/ออสเตรีย ในต้นศตวรรษที่ 20 โดยการ ทำงานของ refractometer เป็นการวัด ดัชนีหักเหของแสง (refractive index) เมื่อเคลื่อนที่ผ่านตัวกลางหนึ่งสู่อีกตัวกลางหนึ่ง เช่น จากอากาศสู่น้ำ จากน้ำสู่คริสตัล (crystal) ทำให้ มุม ความเร็ว (velocity, v) ของแสงแตกต่างกัน

สารละลายที่มีความเข้ม ข้นแตกต่างกันเมื่อแสงส่องผ่านจะเกิด การหักเห และให้ค่าดัชนีหักเหของแสงต่างกันซึ่งจากความสัมพันธ์ดังกล่าว จึงนำมาประยุกต์ใช้วัดค่าความเข้มข้นของสารละลายได้

การวัดค่าดัชนีหักเหของแสง (refractive index) สามารถทำได้ 2 ระบบ คือ

• วัดด้วยการส่องผ่านของแสง (transparent system)
• วัดด้วยการสะท้อนของแสง (reflection system)

ประเภทของ Refractometer ที่ใช้ในอุตสาหกรรมอาหาร

Hand held refractometer เป็น refractometer ที่มีขนาดกะทัดรัด น้ำหนักเบา พกพาได้ ใช้งานง่าย สะดวก รวดเร็ว การทำงานของ hand held refractometer อาศัยหลักการหักเหของแสงใช้วัดความเข้มข้นของสารละลาย

วิธีการ วัดค่า ทำได้โดยการหยดสารละลายที่ต้องการทราบค่าบนแผ่นปริซิม ปิดด้วยแผ่นปิด แล้วส่องมองผ่านช่องในที่มีแสง จะมองเห็นเป็นแถบสี ที่อ่านค่าตัวเลขได้ ตามสเกล ที่เครื่องกำหนดไว้ เช่น เป็นเปอร์เซ็นต์ความเข้มข้น ความเข้มข้นของน้ำตาล น้ำเชื่อม น้ำผลไม้ ที่วัดได้ มีหน่วยเป็น องศาบริกซ์ ( Brix) หรือ อาจเป็นค่าความหนาแน่นของเหลว หรือทั้งสองอย่าง

• ก่อน และหลังการใช้ hand refractometer ควรทำความสะอาดแผ่นปริซึม ซึ่งเป็นบริเวณที่ใช้หยดสารละลายตัวอย่าง ด้วยกระดาษเช็ดเลนส์ ฝุ่นละออง คราบสกปรกบนแผ่นปริซึม มีผลต่อความถูกต้องของค่าที่อ่านได้
• สารละลายที่หยดไม่ควรมีฟองอากาศปน ซึ่งอาจจะมีผลต่อค่าที่อ่านได้
• ค่าที่อ่านได้เป็นค่าที่สัมพันธ์กับความเข้มข้นที่อุณหภูมิที่ระบุไว้ในเครื่องคือ 20 องศาเซลเซียส
